Regulation of the redox potentials of flavodoxins: modification of the flavin binding site

چکیده

Introduction The flavodoxins are a group of small proteins found in micro-organisms and eukaryotic algae that function to transfer electrons at low redox potential between other redox proteins in reactions such as nitrogen fixation and the photosynthetic reduction of NADP’ [ 1-41. They contain a single molecule of FMN bound strongly but non-covalently to the protein. The flavin is reduced in two one-electron steps, first to the neutral semiquinone and then to the hydroquinone anion.

A crystallographic study of Cys69Ala flavodoxin II from Azotobacter vinelandii: structural determinants of redox potential.

Flavodoxin II from Azotobacter vinelandii is a "long-chain" flavodoxin and has one of the lowest E1 midpoint potentials found within the flavodoxin family. To better understand the relationship between structural features and redox potentials, the oxidized form of the C69A mutant of this flavodoxin was crystallized and its three-dimensional structure determined to a resolution of 2.25 A by mole...

Crystallisation of the flavodoxin from Megasphaera eldenii .

Flavodoxins are small flavoproteins (M, 15000-22000), that have been found in several strains of bacteria and algae [ I ] The protein contains one molecule of non-covalently bound FMN and hnctions biochemically as a one-electron carrier between the semiquinone and hlly reduced forms. Under iron-deficient conditions the protein is produced by Megasphaera elsdenii to replace the iron-sulphur prot...

Theoretical determination of the redox potentials of NRH:quinone oxidoreductase 2 using quantum mechanical/molecular mechanical simulations.

NRH:quinone oxidoreductase 2 (NQO2) is a flavoenzyme that catalyzes a one-step two-electron reduction of quinones. During this enzyme catalysis, the 7,8-dimethyl isoalloxazine (flavin) ring of the enzyme-bound cofactor, flavin adenine dinucleotide (FAD), shuttles between reduced and oxidized states as the enzyme passes through multiple cycles of binding/release of alternate substrates. These re...

The Human Thioredoxin System: Modifications and Clinical Applications

The thioredoxin system, comprising thioredoxin (Trx), thioredoxin reductase (TrxR) and NADPH, is one of the major cellular antioxidant systems, implicated in a large and growing number of biological functions. Trx acts as an oxidoreductase via a highly conserved dithiol/disulfide motif located in the active site ( Trp-Cys-Gly-Pro- Cys-Lys-). Different factors are involved in the regulation of T...

Conformational Dynamics of Escherichia coli Flavodoxins in Apo- and Holo-States by Solution NMR Spectroscopy

Flavodoxins are a family of small FMN-binding proteins that commonly exist in prokaryotes. They utilize a non-covalently bound FMN molecule to act as the redox center during the electron transfer processes in various important biological pathways. Although extensive investigations were performed, detailed molecular mechanisms of cofactor binding and electron transfer remain elusive. Herein we r...
